Description：Recombinant human Biotin-Kras[G12A] (GmppNp Loaded) protein with N-terminal 8XHis-TEV, C-terminal Avi tag was purified by Ni affinity and followed by biotinylation and SEC chromatography. The protein was loaded GmppNp, and followed by desalination again. The Biotin-Kras[G12A] (GmppNp Loaded) protein showed good activity in HTRF assay.

Activity Identification：The Biotin-Kras[G12A] (GmppNp Loaded) activity was quantified using a HTRF-based assay. The reaction was performed by incubating the Biotin-Kras[G12A] (GmppNp Loaded) protein, GTP, substrate and the donor and receptor, and keeping the mixture at 25℃ for 60 min, finally measuring the 665nm/620nm fluorescence signal ratio on BMG.
