Description：Recombinant human Biotin-KRAS[G12D] (GDP loaded) protein with N-terminal His tag and C-terminal Avi tag was purified by Ni-NTA affinity and specifically biotinylated at Avi tag in vitro, followed by the His tag was removed using TEV protease. Finally, the protein was loaded with GDP and purified by SEC chromatography. The Biotin-KRAS[G12D] (GDP loaded) protein showed high activity in TR-FRET-based assay.

The reaction was performed by incubating the Biotin-KRAS[G12D](GDP loaded) protein and substrate, the donor and receptor at 25℃ for 60 min, then measuring the 520nm/485nm fluorescence signal ratio on BMG.
