Description：Recombinant human Biotin-KRAS[G12D] (GmppNp loaded) protein with N-terminal His tag and C-terminal Avi tag was purified by Ni-NTA affinity and specifically biotinylated at Avi tag in vitro, followed by the His tag was removed using TEV protease. Finally, the protein was loaded with GmppNp and purified by SEC chromatography. The Biotin-KRAS[G12D] (GmppNp loaded) protein showed high activity in HTRF-based assay.

Activity Identification：The Biotin-KRAS[G12D](GmppNp loaded) activity was quantified using a HTRF-based assay. The reaction was performed by incubating the Biotin-KRAS[G12D](GmppNp loaded) protein, GTP and substrate at 25℃ for 60 min, then adding the donor and receptor, and keeping the mixture at 25℃ for 60 min, finally measuring the 665nm/620nm fluorescence signal ratio on BMG.
