Description：Recombinant human Biotin-NRAS[Q61L] (GmppNp loaded) protein with N-terminal His tag and C-terminal Avi tag was purified by Ni-NTA affinity and specifically biotinylated at Avi tag in vitro, and followed by desalting and the protein was loaded with GmppNp. Finally, followed by SEC chromatography. The Biotin-NRAS[Q61L] (GmppNp Loaded) protein showed high activity in HTRF assay.

Activity Identification：The Biotin-NRAS[Q61L] (GmppNp loaded) activity was quantified using a HTRF-based assay. The reaction was performed by incubating the Biotin-NRAS[Q61L] (GmppNp loaded) protein, GTP and substrate at 25℃ for 60 min, then adding the donor and receptor, and keeping the mixture at 25℃ for 60 min, finally measuring the 665nm/620nm fluorescence signal ratio on BMG.
