Description：Recombinant human Biotin-KRAS[G12C/H95D] (GDP loaded) protein with N-terminal His tag and C-terminal Avi tag was purified by Ni-NTA affinity and specifically biotinylated at Avi tag in vitro, followed by His tag removal with TEV protease. Finally, the protein was further purified by SEC chromatography and loaded with GDP. The Biotin-KRAS[G12C/H95D] (GDP Loaded) protein showed high activity in TR-FRET assay.

Activity Identification：The Biotin-KRAS[G12C/H95D] (GDP Loaded) activity was quantified using TR-FRET-based assay. The reaction was performed by incubating the Biotin-KRAS[G12C/H95D](Loaded GDP) protein and substrate, the donor and receptor at 25℃ for 180 min, then measuring the 520nm/485nm fluorescence signal ratio on BMG.
