Description：Recombinant human TOP1 protein with N-terminal 6x His tag and C-terminal Avi tag was performed biotinylation in vivo. Recombinant human Biotin-TOP1 protein was purified by Ni-NTA affinity and followed by SEC chromatography. The Biotin-TOP1 protein showed high activity in DNA relaxation assay.

Activity Identification：The Biotin-TOP1 activity was detected by DNA relaxation assay. The reaction was performed by incubating the Biotin-TOP1 protein and plasmid TR-pBR322 at 37℃ for 10 min, then adding 10% SDS and protease K working solution, and mix them at 37℃ for 30 min and adding the mixture of chloroform : isoamylol (24:1) and invert and mix well, and then remove the supernatant after standing then the agarose gel electrophoresis and band analysis.
