Description:Recombinant human Biotin-STAT6 (phosphorylation) protein with N-terminal His tag and C-terminal Avi tag was purified by Ni-NTA affinity and followed by SEC chromatography. The protein was activated by JAK1, biotinylated and enzymatically cleaved in vitro. Subsequently, GST affinity and SEC chromatography were performed. The Biotin-STAT6 (phosphorylation) protein showed good activity in TR-FRET assay.

Activity Identification:The Biotin-STAT6 (phosphorylation) activity was quantified using TR-FRET-based assay. The reaction was performed by incubating the Biotin-STAT6 (phosphorylation) protein and substrate, the donor and receptor at 25℃ for 60 min, then measuring the 520nm/485nm fluorescence signal ratio on BMG.
