Description：Recombinant human KEAP1[S172A] protein with N-terminal 6x His tag was purified by Ni-NTA affinity and followed by His-tag removal with TEV protease and final purification by SEC chromatography. SEC-HPLC result showed that the KEAP1[S172A] protein was a dimer in the solution. The KEAP1[S172A] protein showed high activity in nanoDSF assay.

Activity Identification：The KEAP1[S172A] activity was quantified using nanoDSF assay. The reaction was performed by incubating the KEAP1[S172A] protein and 1%DMSO at 25℃ for 15 min, and then on PrometheusTM NT.48 heating the reaction mixture with the temperature range from 20℃ to 95℃, and real time measuring 350/330 nm fluorescence signal ratio.
