Activity Identification:The Biotin-STAT6[Y616W] activity was quantified using using a SPR-based assay. The Biotin-STAT6[Y616W] protein was immobilized onto the CM5 chip. The varying concentration of STAT6-IN-3 was injected, and flowed over onto the Biotin-STAT6[Y616W] protein surface. And the response signal was recorded real time. The kinetics fitting was as follows with a KD value of ~ 2.54x10-8 M.
