Description:Recombinant human PIK3CA[E545K] with C-terminal 8x His tag was co-expressed with PIK3R1 protein with N-terminal Flag tag and C-terminal Avi tag. Recombinant human Biotin-PIK3CA[E545K]/PIK3R1 protein was purified by Ni-NTA affinity and biotinylation and followed by SEC chromatography. The Biotin-PIK3CA[E545K]/PIK3R1 protein showed high activity in ADP-Glo assay.

Activity Identification:The Biotin-PIK3CA[E545K]/PIK3R1 activity was detected using Ultra Luminescence technology. The reaction was performed by incubating the Biotin-PIK3CA[E545K]/PIK3R1 protein and substrate at 25℃ for 60 min, then adding ADP-Glo reagent and reading RLU signal with BMG.
