Activity Identification：The Biotin-PKACα activity was quantified using a HTRF-based assay. The reaction was performed by incubating the Biotin-PKACα protein, ATP and substrate at 25℃ for 60 min, then adding the donor and receptor, and keeping the mixture at 25℃ for 60 min, finally measuring the 665nm/620nm fluorescence signal ratio on BMG.
