Description:Recombinant human METTL3 protein with N-terminal Flag tag was co-expressed with recombinant human METTL14 with N-terminal 6x His tag. Recombinant human METTL3/METTL14 protein was purified by Ni-NTA affinity and followed by SEC chromatography. The METTL3/METTL14 protein showed high activity in HTRF assay.

Activity Identification:The METTL3/METTL14 activity was detected using HTRF technology. The reaction was performed by incubating the METTL3/METTL14 protein and Bio-METTL3/ METTL14-RNA&SAM at 25℃ for 60 min, adding the YTHDF1 and beads at 25℃ for 60 min, then reading ratio 665/620 with BMG.
