Activity Identification：The BRD4 (BD2) activity was quantified using a SPR-based assay. The BRD4 (BD2) protein was immobilized onto the CM5 chip. The varying concentration of (+)-JQ-1 was injected, and flowed over onto the BRD4 (BD2) protein surface. And the response signal was recorded real time. The kinetics fitting was as follows with a kD value of ~ 1.46x10-7 M.
