Description：Recombinant human CRBN with N-terminal 6x His and Avi tag was co-expressed with recombinant human DDB1 protein and performed in vivo biotinylation simultaneously. Recombinant human Biotin-CRBN/DDB1 protein was purified by Ni-NTA affinity and followed by desalting. The Biotin-CRBN/DDB1 protein showed good activity in TR-FRET assay.

Activity Identification：The Biotin-CRBN/DDB1 activity was detected using TR-FRET technology. The reaction was performed by incubating the Biotin-CRBN/DDB1 protein and Thalidomide-BODIPY, adding the beads and keeping the mixture at 25℃ for 60 min, then reading ratio 520/490 with BMG.
