Description：Recombinant human CRBN with N-terminal 6x His and Avi tag was co-expressed with recombinant human DDB1 protein. Recombinant human CRBN/DDB1 protein was biotinylated in vivo. The recombinant human CRBN/DDB1 protein was purified using Ni-NTA affinity, followed by His-tag removal with TEV protease and final purification by size exclusion chromatography. The Biotin-CRBN/DDB1 protein showed good activity in TR-FRET assay.

Activity Identification：The Biotin-CRBN/DDB1 activity was quantified using TR-FRET-based assay. The reaction was performed by incubating the Biotin-CRBN/DDB1 protein and substrate, the donor and receptor at 25℃ for 60 min, then measuring the 520nm/490nm fluorescence signal ratio on BMG.
